Pantoprazole
medication Under reviewA proton pump inhibitor (PPI) used to treat conditions involving excessive stomach acid, such as GERD.
Research summary
Pantoprazole has been evaluated in over one hundred clinical trials, including randomized controlled trials, cohort studies, and long‑term extension studies in adults. The overall research consensus is that pantoprazole provides effective and durable gastric acid suppression, leading to high rates of healing in erosive esophagitis and substantial improvement in GERD symptoms and patient‑reported quality of life in otherwise healthy subjects. Comparative studies indicate that pantoprazole is at least as effective as other proton pump inhibitors and generally more effective than histamine‑2 receptor antagonists for both acute treatment and maintenance therapy of GERD. Its tolerability profile in healthy adults is favorable, with most adverse events being mild and transient gastrointestinal or headache‑type symptoms. Long‑term observational data and regulatory reviews highlight potential risks associated with chronic PPI therapy, including nutrient malabsorption, alterations in bone health, and increased susceptibility to certain infections, but these are typically infrequent and more strongly associated with prolonged high‑dose use.
