Human research on phenindione is almost entirely in the context of therapeutic anticoagulation rather than use as a general supplement in healthy individuals. Comparative studies from the mid‑20th century evaluated phenindione against other vitamin K antagonists and found broadly similar anticoagulant efficacy and ability to achieve therapeutic prothrombin inhibition when appropriately dosed and monitored. Detailed mechanistic work in human subjects described multiphasic effects on the coagulation system, confirming that phenindione reduces activity of vitamin K–dependent clotting factors and alters various stages of the clotting cascade. There are no controlled trials in healthy volunteers evaluating phenindione for general health optimization, performance, longevity or other typical supplement‑style outcomes. Instead, the evidence base focuses on its pharmacodynamics, dosing strategies, and comparative safety and efficacy as a prescription anticoagulant in patients with cardiovascular and thromboembolic disease.
